Plant Preservation Mixture (PPMTM) is a robust broad-spectrum biocide formulated for use in plant tissue culture. PPMTM targets bacteria and fungi in plant tissue culture growth media as well as contaminated tissue. It affects key enzymes in the Krebs cycle and in the Electron Transport Chain. Depending on the dose and the level of contamination, PPM is a biocidal component in plant culture medium. In addition, it may also function as a biostatic compound as a preventative measure. When diluted with plant growth media it is effective as a microbiocide (i.e. bactericide and fungicide) against non-human health pathogenic organisms component of liquid or semi-solid plant culture media.

Plant Preservative Mixture, also known as PPM, is a solution that is used to preserve plant tissues for scientific research or other purposes. It is typically made up of several chemicals that help to prevent the tissues from deteriorating or becoming contaminated during storage or transportation.

There are many different formulations of PPM, and the specific composition of the mixture can vary depending on the specific needs of the research or application. Common ingredients in PPM formulations include chemicals that help to inhibit the growth of microbes (such as antibiotics or antimicrobial agents), protect against oxidative damage (such as antioxidants), and maintain the structural integrity of the plant tissues (such as tissue fixation agents).

PPM is commonly used in research and diagnostic laboratories to preserve plant tissues for analysis or other purposes. It is also used in a variety of other applications, including horticulture and agriculture. It is important to follow the manufacturer's instructions and safety precautions when handling PPM, as some of the chemicals it contains may be toxic or hazardous.

PPMTM is effective for most seed baring plants - angiosperm, as well as gymnosperm,- however, it is not recommended for use in ferns, mosses, algae and aquatic plants. Optimization may be required to maximize potency. While PPM is an excellent tool in the prevention and elimination of culture contamination it is not a substitute for asceptic laboratory techniques and appropriate air handling systems are recommended.

Protocol for Plant Preservative Mixture, PPM

Plant Preservative Mixture (PPM), is used as a soft antibiotic for plant tissue culture, callus propagation.

PPM™ preventing microbial airborne, waterborne and endogenous contamination.

PLANT PRESERVATIVE MIXTURE  PPM Properties

PPM™ is a heat stable preservative/biocide which can be used to effectively prevent or reduces microbial contamination in plant tissue culture.

At optimum doses, PPM™, which stands for Plant Preservative Mixture™, is an extremely effective Preservative/Biocide, yet does not impair in vitro seed germination, callus proliferation and callus regeneration.
PPM™ prevents the germination of both bacteria and fungi spores. It is heat stable and therefore can be autoclaved with the media.

PPM™ can be, and should be used as a standard ingredient in plant tissue culture media, and is also substantially less expensive than commonly used antibiotics.
While PPM™ was principally designed to inhibit airborne contamination, waterborne contamination and contamination introduced from human contact, it can also -- in many cases -- be used to reduce endogenous contamination.

Despite the most stringent use of sterile techniques, the contamination of plant cell and plant tissue cultures remain a persistent problem that can result in losses ranging from small number of cultures to the loss of whole batches.
